Focusing on the construction of organoid systems and their application in biomedical research, core scientific questions encompass stem cell fate determination, organ homeostasis regulation, tumour classification, and drug therapy. Specifically, utilising organoid models of colorectal, pancreatic and breast cancers, combined with mouse models, multi-omics technologies and signalling pathway interventions, we aim to elucidate mechanisms governing tissue homeostasis maintenance and dysfunction during disease progression. We further investigate the pivotal role of cellular interactions and fate regulation in tumourigenesis and progression. Beyond technological innovation, the research endeavours to provide robust theoretical foundations and translational solutions for clinical disease diagnosis and treatment.
